354 Unexpected elasticity-boosting properties of a traditional ayurvedic plant

The loss of elasticity is a characteristic feature skin aging. Indeed, elastic fibers are poorly renewed and degraded much quicker than synthesized. To compensate for this imbalance, unique extract from the traditional ayurvedic plant Murraya koenigii was developed. First, proteomic study combined with bioinformatics, revealed that monolayer cultures human dermal fibroblasts (HDF) treated M. expressed increased levels proteins involved in maintenance integrity extracellular matrix. More specifically, we demonstrated able to significantly increase synthesis situ deposition elastin (+43%), fibrillin-1 (+22%) fibulin-5 (+62%) HDF. As both epidermis dermis required proper fiber formation, these results were further confirmed full-thickness 3D reconstructed model, (+103% elastin, +35% extract). also reduced degradation by inhibiting MMP-12 elastase activity (-27%). control structural functional quality neo-synthesized presence extract, developed scaffold-free spheroid microtissue, which network characterized using two-photon autofluorescence (2PAF) imaging. Based on 2PAF image analysis, microtissue exhibited denser more mature network. Regarding biomechanical properties, showed atomic force microscopy (AFM), 3-fold decrease relative stiffness after treatment koenigii. AFM analyses allowed draw correlation map apparent modulus deposit density, elastin-rich areas less stiff. conclude, displays undeniable boosting highlighted at molecular, cellular tissue levels, appears as powerful anti-aging ingredient.
